preparation
1L sesame oil feet (965.8g) were measured, 4L of anhydrous ethanol was added, stirred at 30℃ for 5h, left for stratification, superclear liquid was taken, and rotary evaporation was performed to obtain sesamin enrichment. Adding anhydrous methanol to sesamin enrichment, the solid-liquid ratio of sesamin enrichment and anhydrous methanol was 1∶4 (kg/L), and leach at 55℃ for 3h to obtain the extracted solution. Activated carbon was added to the leach solution, the amount of activated carbon was 1% of the quality of the leach solution, stirred for 0.5h, and then filtered to obtain sesamin extract solution. The sesamin extract was slowly cooled to -10℃, the sesamin was crystallized, and the Chemicalbook was crystallized for 2h, and the crude sesamin was filtered. The crude sesamin was filtered by adding petroleum into the crude sesamin, the solid-liquid ratio of the crude sesamin and petroleum was 1∶1 (kg/L), and the crude sesamin was filtered by mixing at 35℃ for 0.5h. The crude sesamin product was dissolved in anhydrous ethanol, and the solid-liquid ratio of the crude sesamin product and anhydrous ethanol was 1:30 (kg/L). The product was crystallized and purified at -5℃ for 3 hours. Under this condition, after two times of crystallization and vacuum drying, the sesamin product 2.16g was obtained.

Content determination
A method for determining sesamin content in sesamin seeds includes the following steps: (1) preparation of sesamin standard solution: weighing 10mg sesamin standard, adding anhydrous methanol to dissolve and dilute to 50mL, and then filtering with 0.45μm filter membrane to obtain 0.2mg/mL sesamin standard solution; (2) Drawing sesamin standard curve: Take 20, 10, 5, 4, 2, 0.8mL of sesamin standard solution obtained in step (1), add 0, 10, 15, 16, 18, 19.2mL of anhydrous methanol, and shake well to prepare the sesamin standard methanol solution with gradient concentrations of 200, 100, 50, 40, 20, 8ug/mL, respectively. Chromatographic analysis of the above solutions was carried out by using liquid chromatograph, and the standard curve of sesamin was drawn according to the measured results (see Figure 1 for the liquid chromatographic diagram of sesamin standard solution), and the linear relationship equation between sesamin content and peak area was obtained: A=587.525675x-4.53158Chemicalbook2, where A is peak area and x is sesamin content; (3) Prepare the sample solution to be tested: Yuzhi No. 11 sesame seed was used as the sample to be tested, the sesame seed to be tested was crushed for 30s with a universal grinder, and then screened with a 0.35μm sieve. The obtained sesame powder sample was 0.2g, and the concentration of 100% ethanol was added to 5mL; (4) Ultrasonic treatment and filtration: the mixed solution obtained in step (3) is subjected to ultrasonic treatment at 30 ~ 60℃ for 20 ~ 50min, and then 0.45μm filtration membrane is used to filter the ultrasonic treated solution; The conditions for ultrasonic treatment are as follows: a CNC ultrasonic cleaner with ultrasonic frequency of 40KHZ and model KQ3200DE is used for ultrasonic treatment at ultrasonic power of 70-100W. (5) Detection: Take 9μl of filtrate obtained in step (4), use liquid chromatograph to sample the filtrate for detection, and calculate the content of sesamin in sesame seeds to be measured according to the standard curve of sesamin and the linear relationship equation obtained.
